How to Calculate Assay Percentage in Pharmaceutical Analysis

Assay calculation is one of the most fundamental calculations in pharmaceutical analysis. It tells you what percentage of the labelled amount of active ingredient is actually present in your sample โ€” whether you are testing a tablet, capsule, injection, or bulk API.

Despite being routine, it is also a source of errors. This guide walks through the correct formula, explains each variable, and gives a fully worked example.


The Assay Formula

For HPLC assay against an external reference standard, the standard formula is:

Assay (%) = (AT ร— WS ร— PS ร— DT) รท (AS ร— WT ร— DS) ร— 100

When test and standard are prepared at the same concentration with the same dilution scheme, DT and DS cancel out and the formula simplifies to: Assay (%) = (AT ร— WS ร— PS) รท (AS ร— WT) ร— 100


Worked Example

Given values:

AT (sample peak area) = 95,432

AS (standard peak area) = 96,110

WS (standard weight) = 50.24 mg

WT (sample weight) = 50.00 mg

PS (standard purity) = 99.2%

Same dilution for both (DT = DS, they cancel)

Calculation:

Assay = (95,432 ร— 50.24 ร— 99.2) รท (96,110 ร— 50.00) ร— 100

= (95,432 ร— 50.24 ร— 99.2) รท (4,805,500) ร— 100

= 475,537,715 รท 4,805,500 ร— 100

= 99.0%

An assay of 99.0% is within the typical acceptance criteria of 98.0โ€“102.0% for pharmaceutical products. This result would pass.
