Dwell (gradient delay) volume is the total volume between the point where solvents mix and the column inlet โ including mixer, tubing, and injector volume. It causes a delay between the programmed gradient and what the column actually experiences.
Why does dwell volume matter for method transfer?
Different HPLC/UHPLC systems have different dwell volumes. A method transferred from a system with high dwell volume to one with low dwell volume (or vice versa) can show shifted retention times unless the gradient program is adjusted or an isocratic hold is added to compensate.
